چکیده

Objective: The dysregulation of metabolism is a hallmark cancer. Enzymes tricarboxylic acid (TCA) cycle have key role in pathogenesis carcinogenesis as oncometabolits. variants fumarate hydratase (FH) gene been linked to sporadic tumors and familial tumor propensity. To asses the prevalence FH endometrial carcinoma (EC) uterine leiomyomas (UL). Material Methods: This prospective study included 58 patients with EC 44 UL. were analyzed DNA samples collected from peripheral circulation. All exons replicated by polymerase chain reaction utilizing specific primers including intron-exon boundaries than was sequenced Sanger method. Results: Only two detected different patients. c.927G>A (p.Pro309Pro) c.63C>T (p.Ala21Ala) observed UL, respectively. (p.Pro309=) (p.Ala21=) are rare inside overall population allele frequency ranges 0.02428, 0.000014 Conclusion: contribution genes encoding TCA enzymes cells has provided understanding fundamental on metabolic alterations carcinogenesis. Further studies needed discover novel mutations identify appropriate treatment.

Distinct expression profile in fumarate-hydratase-deficient uterine fibroids.

Defects in mitochondrial enzymes predispose to severe developmental defects as well as tumorigenesis. Heterozygous germline mutations in the nuclear gene encoding fumarate hydratase (FH), an enzyme catalyzing the hydration of fumarate in the Krebs tricarboxylic acid cycle, cause hereditary leiomyomatosis and renal cell cancer; yet the connection between disruption of mitochondrial metabolic pat...

Germline fumarate hydratase mutations and evidence for a founder mutation underlying multiple cutaneous and uterine leiomyomata.

Multiple cutaneous and uterine leiomyomata syndrome (MCL) is an autosomal dominant disease characterized by the presence of concurrent benign tumors of smooth muscle origin (leiomyoma) in the skin and uterus of affected females, and in the skin of affected males. MCL can also be associated with type II papillary renal cell cancer (HLRCC). The genetic locus for MCL and HLRCC was recently mapped ...

Germline fumarate hydratase mutations in families with multiple cutaneous and uterine leiomyomata.

Germline mutations in the fumarate hydratase gene (FH) predispose to multiple cutaneous and uterine leiomyoma syndrome (MCL) and MCL associated with renal cell cancer. MCL is inherited in an autosomal dominant pattern, manifesting as skin leiomyoma and uterine fibroids in affected individuals. Fumarate hydratase, a component of the tricarboxylic acid cycle, acts as a tumor suppressor gene in th...

A Case of Reed Syndrome with a Novel Mutation in the Fumarate Hydratase Gene

Reed syndrome is a heritable cancer predisposition syndrome that can easily be missed due to its simple presentation of tender red papules. We present a young female with a history of uterine fibroids who presented to the dermatology clinic with several painful pink papules that had been previously evaluated by multiple physicians. Biopsy results were diagnostic for cutaneous leiomyomas, raisin...
